PLANT REGENERATION FROM IMMATURE EMBRYO-DERIVED CALLUS OF BARLEY (Hordeum vulgare L.): EFFECT OF AUXINS AND CYTOKININS
ثبت نشده
چکیده
Embryogenic callus was induced from culturing immature embryos of three barley cvs. Dissa, Golden Promise and Igri. Maximum embryogenic callus formation was achieved by culturing immature embryos on CC and LS medium containing 2 mg/litre and 2.5 mg/litre 2,4dichlorophenoxyacetic acid (2,4-D) respectively. Embryogenic calli were frequently produced distinguished somatic embryos on these medium. Highest frequency of somatic embryo’s germination was occurred on MS medium supplemented with 0.5 mg/litre kinetin and 3% glucose in cv. Dissa. This medium also induced regeneration in the calli of cv. Golden Promise and Igri. The age of callus also influenced regeneration with high frequencies occurring after 6 wk in culture. Subsequently, plant regeneration was achieved by transferring germinated somatic embryos on hormone-free MS medium. It indicated that plant regeneration was achieved via somatic embryogenesis and organogenesis. Plants regenerated from immature embryos were grown to maturity in soil.

Sorghum is a wonder crop from physiological point of view. It is the most important cereal crop, after rice and wheat. The number of reports describing the use of transgenic Sorghum for basic studies in Biotechnology is still limited when compared to other crops. In one hand, the success of the transformation techniques is mainly dependent upon the availability of optimal protocols for highly ...
متن کاملIn vitro Plant Regeneration from the Callus of Shoot Tips in Cotton (Gossypium hirsutum L. cv. SVPR 2)
A high efficiency shoot organogenesis from callus cultures and plant establishment protocol has been developed for cotton (Gossypium hirsutum L. cv SVPR-2) through shoot tip. After 6 weeks of culture, green-compact-nodular organogenic callus induction was observed on the medium fortified with (MS) basal salts, B5 vitamins, 2.0 mgl-1 of benzylaminopurine (BAP), 1.0 mgl-1 kinetin (KN), 30 gl-1 gl...
متن کاملCallus Induction and Somatic Embryogenesis from Maize Mature Embryos (zea Mays L.)
Ten maize lines prevalently used by breeders in the maize program were screened for their ability to form type II callus. Mature seeds were used as explant for in vitro culturing. Inbred lines showed high embryogenic response and formed friable, type II calli. And also exhibited successful plant regeneration. Various culture media compositions based on Murashige and Skoog (1962) salts thus inve...

Eclipta alba is a medicinal herb, also known as Bhringraj belonging to family Asteraceae. The plant has hepatoprotective, antimicrobial and hair growth promoting properties. The effect of various phytohormones on in-vitro callus induction and shoot regeneration in Eclipta alba from leaf explant was evaluated. The sterilized explants were inoculated on MS medium supplemented with different conce...

Einkorn (Triticum monococcum L.) is A-genome diploid wheat that has a potential to become a useful model for understanding the biology and genomics in Triticeae. Unfortunately, the application of modern technologies such as genetic engineering, RNAi-based gene silencing and genome editing is not available for einkorn as there is no efficient in vitro tissue culture and plant regeneration system...
